Depuis 2017, elle dirige le d\u00e9partement de biologie et est une experte reconnue en bioanalyse de macromol\u00e9cules, en dosage d&rsquo;ADA et <a href=\"https:\/\/kymos.com\/fr\/services\/services-precliniques-et-cliniques\/activite-enzymatique\/\" data-wpel-link=\"internal\">en mesure de l&rsquo;activit\u00e9 enzymatique<\/a> . Avec le Dr Busch, directeur des op\u00e9rations chez Eleva, elle partage son expertise dans le domaine prometteur des enzymes th\u00e9rapeutiques. Lisa Hentschel revient sur son parcours depuis 2014.   <\/span><\/p>\n<p style=\"padding-left: 40px;\"><span style=\"font-size: 14pt;\"><em>\u00ab Il y a huit ans, Eleva, alors encore Greenovation, nous a contact\u00e9s avec un probl\u00e8me pr\u00e9cis pour lequel personne n&rsquo;avait pu les aider jusque-l\u00e0. Au lieu d&rsquo;une quantification non sp\u00e9cifique de leur enzyme dans le plasma, ils souhaitaient conna\u00eetre la quantit\u00e9 d&rsquo;enzyme active pr\u00e9sente apr\u00e8s administration. \u00bb<\/em><\/span><\/p>\n<p><span style=\"font-size: 14pt;\">Le Dr Busch revient \u00e9galement sur la situation dans laquelle se trouvait Eleva \u00e0 l&rsquo;\u00e9poque et explique pourquoi il a contact\u00e9 Prolytic pour leur faire part de leur demande&nbsp;:<\/span><\/p>\n<p style=\"padding-left: 40px;\"><span style=\"font-size: 14pt;\"><em>\u00ab Certaines des maladies g\u00e9n\u00e9tiques humaines les plus graves sont dues \u00e0 un d\u00e9ficit enzymatique. Chez Eleva, nous avons r\u00e9solu ce probl\u00e8me gr\u00e2ce \u00e0 un outil incroyablement efficace&nbsp;: les bryophytes, plus commun\u00e9ment appel\u00e9es mousses. Nous en extrayons l\u2019alpha-galactosidase et d\u2019autres enzymes que nous utilisons dans nos th\u00e9rapies enzymatiques de substitution. Pour \u00e9valuer l\u2019efficacit\u00e9 de nos enzymes, nous devons d\u00e9terminer non seulement leur concentration sanguine, mais aussi leur activit\u00e9. Malheureusement, nous n\u2019avons trouv\u00e9 aucun prestataire capable de r\u00e9aliser ces analyses selon les BPL et nous nous sommes finalement tourn\u00e9s vers Prolytic. \u00bb<\/em><\/span><\/p>\n<p><span style=\"font-size: 14pt;\">Lisa Hentschel poursuit le r\u00e9cit&nbsp;: <em>\u00ab&nbsp;C\u2019est ainsi qu\u2019est n\u00e9 notre syst\u00e8me d\u2019analyse sp\u00e9cialis\u00e9 pour la mesure de l\u2019activit\u00e9 enzymatique. Une m\u00e9thode \u00e9tait disponible chez Eleva, mais sa validation selon les BPL s\u2019av\u00e9rait complexe, car il n\u2019existait aucune directive pour ce cas particulier. Par cons\u00e9quent, mon \u00e9quipe et moi avons \u00e9labor\u00e9 un sch\u00e9ma de validation conforme aux recommandations de validation des m\u00e9thodes bioanalytiques pour l\u2019industrie en mati\u00e8re de dosage de la liaison de ligands (LBA).&nbsp;\u00bb<\/em><\/span><\/p>\n<p><span style=\"font-size: 14pt;\">Depuis, le d\u00e9partement de biologie a d\u00e9velopp\u00e9 et valid\u00e9 des tests d&rsquo;activit\u00e9 pour de nombreuses enzymes provenant de diff\u00e9rents clients, \u00e9largissant et professionnalisant ainsi ses connaissances et son exp\u00e9rience. Cependant, l&rsquo;\u00e9quipe de Lisa Hentschel ne se limite pas \u00e0 l&rsquo;activit\u00e9 enzymatique&nbsp;: l&rsquo;analyse bioanalytique de macromol\u00e9cules par ELISA fait partie int\u00e9grante de son domaine d&rsquo;expertise, au m\u00eame titre que la d\u00e9termination des anticorps anti-m\u00e9dicament (ADA). Depuis l&rsquo;int\u00e9gration de Prolytic au sein du groupe Kymos, un plus grand nombre de tests sont disponibles, comme l&rsquo;explique Lisa Hentschel&nbsp;:  <\/span><\/p>\n<p style=\"padding-left: 40px;\"><span style=\"font-size: 14pt;\"><em>\u00ab Nos clients nous demandaient r\u00e9guli\u00e8rement des services analytiques compl\u00e9mentaires, que nous ne pouvions pas proposer en tant que petite CRO. Gr\u00e2ce \u00e0 notre collaboration avec nos coll\u00e8gues de Barcelone, nous pouvons d\u00e9sormais fournir des tests de neutralisation et de liaison cellulaire, en plus de nos services ADA. Sans oublier les possibilit\u00e9s offertes par notre partenaire GMP, le d\u00e9partement \u00ab&nbsp;Contr\u00f4le Qualit\u00e9 des Produits Biologiques et Biosimilaires&nbsp;\u00bb. Cela nous permet, au sein du Groupe Kymos, d&rsquo;accompagner nos clients depuis les premi\u00e8res phases de d\u00e9veloppement jusqu&rsquo;\u00e0 la production, la commercialisation et au-del\u00e0.
