L&rsquo;harmonisation des exigences relatives \u00e0 ces m\u00e9thodes a d\u00e9but\u00e9 en 1990 lors des r\u00e9unions conjointes de l&rsquo;AAPS et de la FDA \u00e0 Crystal City. En 2000, la r\u00e9union de Crystal City II a jet\u00e9 les bases des lignes directrices de la FDA de 2001 sur la validation des m\u00e9thodes bioanalytiques. En 2011, l&rsquo;EMA a publi\u00e9 ses propres lignes directrices sur la validation des m\u00e9thodes bioanalytiques, fournissant des recommandations pour la validation des m\u00e9thodes appliqu\u00e9es \u00e0 la mesure des concentrations de m\u00e9dicaments dans les matrices biologiques obtenues lors d&rsquo;\u00e9tudes toxicocin\u00e9tiques animales et \u00e0 toutes les phases des essais cliniques. Ces lignes directrices ont harmonis\u00e9 la validation des m\u00e9thodes bioanalytiques impliqu\u00e9es dans les \u00e9tudes pr\u00e9cliniques, cliniques et de bio\u00e9quivalence. Toutefois, les exigences pour satisfaire \u00e0 ces lignes directrices sont co\u00fbteuses et chronophages, et ne correspondent pas aux besoins de l&rsquo;industrie lorsque les analyses bioanalytiques sont con\u00e7ues pour prendre des d\u00e9cisions de faible pertinence lors des premi\u00e8res phases du d\u00e9veloppement des m\u00e9dicaments et lorsqu&rsquo;elles sont utilis\u00e9es dans des matrices sp\u00e9cifiques.     <\/span><\/p>\n<div class=\"blog-content\">\n<div class=\"field field-name-body field-type-text-with-summary field-label-hidden\">\n<div class=\"field-items\">\n<div class=\"field-item even\">\n<div>\n<p><span style=\"font-size: 14pt;\">Depuis 2006, le concept d&rsquo;approche <strong>adapt\u00e9e \u00e0 l&rsquo;usage<\/strong> s&rsquo;est int\u00e9gr\u00e9 aux discussions bioanalytiques suite \u00e0 la proposition d&rsquo;une approche par paliers pour la validation des m\u00e9thodes bioanalytiques lors de l&rsquo;\u00e9tude Crystal City III. Cette approche par paliers propose diff\u00e9rents niveaux de <strong>validation scientifique<\/strong> comme alternative \u00e0 l&rsquo;application int\u00e9grale des principes de validation r\u00e9glementaire \u00e9tablis. Plus flexible, elle permet de distinguer quatre niveaux d\u00e9croissants de performance et d&rsquo;\u00e9valuation de la m\u00e9thode, en fonction de la finalit\u00e9 des donn\u00e9es bioanalytiques&nbsp;: 1) Validation, destin\u00e9e aux \u00e9tudes r\u00e9glementaires, 2) Qualification, 3) Recherche et 4) Criblage.      <\/span><\/p>\n<p><span style=\"font-size: 14pt;\">Plus r\u00e9cemment, des repr\u00e9sentants de l&rsquo;industrie (au sein du Forum europ\u00e9en de bioanalyse) ont propos\u00e9 de red\u00e9finir cette approche par \u00e9tapes sous l&rsquo;angle de <strong>la validation scientifique<\/strong> . L&rsquo;objectif serait de mettre l&rsquo;accent sur la qualit\u00e9 des donn\u00e9es g\u00e9n\u00e9r\u00e9es par ces flux de travail de validation alternatifs, tout en respectant l&rsquo;objectif d&rsquo;optimisation des ressources et des efforts scientifiques. Ils ont d\u00e9fini une application pratique de ce concept dans cinq domaines cl\u00e9s du soutien bioanalytique&nbsp;: 1) la quantification des m\u00e9tabolites en phase pr\u00e9coce de d\u00e9veloppement (conform\u00e9ment aux recommandations de Crystal City III), 2) l&rsquo;analyse d&rsquo;urine \u00e0 tous les stades du d\u00e9veloppement, 3) l&rsquo;analyse tissulaire (homog\u00e9nat) \u00e9galement \u00e0 tous les stades du d\u00e9veloppement, 4) les \u00e9tudes cliniques en phase pr\u00e9coce de d\u00e9veloppement du m\u00e9dicament, et 5) les \u00e9tudes pr\u00e9cliniques BPL non pivotales.      <\/span><\/p>\n<p><span style=\"font-size: 14pt;\">Le plan de qualification pour une validation scientifique <strong>adapt\u00e9e \u00e0 l&rsquo;objectif<\/strong> doit \u00eatre d\u00fbment justifi\u00e9 afin de r\u00e9pondre aux objectifs de l&rsquo;\u00e9tude avec un niveau ad\u00e9quat de fiabilit\u00e9 et de qualit\u00e9 des
