Els oligonucle\u00f2tids antisentit (ASO), els ARN d&#8217;interfer\u00e8ncia petits (siRNA), els apt\u00e0mers, els terap\u00e8utics d&#8217;ARN missatger (mRNA) i altres modalitats basades en \u00e0cids nucleics progressen r\u00e0pidament a trav\u00e9s del desenvolupament de f\u00e0rmacs, impulsant la necessitat de m\u00e8todes bioanal\u00edtics robustos capa\u00e7os de caracteritzar amb precisi\u00f3 aquestes mol\u00e8cules complexes (1,2). <\/span><\/p>\n<p><span style=\"font-size: 14pt;\">En comparaci\u00f3 amb les petites mol\u00e8cules tradicionals, els terap\u00e8utics d&#8217;oligonucle\u00f2tids presenten reptes anal\u00edtics \u00fanics. La seva mida molecular relativament gran, l&#8217;alta polaritat i la susceptibilitat a la degradaci\u00f3 mediada per nucleases compliquen l&#8217;extracci\u00f3 i la quantificaci\u00f3 de matrius biol\u00f2giques. A difer\u00e8ncia dels assajos ELISA d&#8217;hibridaci\u00f3, la LC-MS\/MS identifica directament els analits segons la seva massa molecular i els patrons de fragmentaci\u00f3 caracter\u00edstics, permetent una quantificaci\u00f3 precisa del compost original juntament amb la discriminaci\u00f3 de metab\u00f2lits estretament relacionats. A difer\u00e8ncia dels m\u00e8todes basats en hibridaci\u00f3, la LC-MS\/MS proporciona una detecci\u00f3 molecular directa, permetent no nom\u00e9s una quantificaci\u00f3 precisa sin\u00f3 tamb\u00e9 la confirmaci\u00f3 estructural de l&#8217;analit.
